Descriptions : Turkish Yapılan bu çalışma ile Çukurova Bölgesinde F.graminearum ve F.culmorum'un varlığını mikolojik ve moleküler yöntemlerle belirlemek ve bölgemizde yetiştirilen Adana 99, Seri 82, ve Genç 99 buğday (Triticum aestivum L.) çeşitlerinin embriyogenik kallusları kullanılarak, in vitro seleksiyon ile Buğday Başak Yanıklığı Hastalığı'na karşı dayanıklı bitkiler elde etme olanakları araştırılmıştır. Survey çalışmaları sonucunda bölgemizde yetiştirilen buğday çeşitlerinde Buğday Başak Yanıklığı Hastalığı'na neden olan F.graminearum ve F. culmorum'un varlığı belirlenmiş ve bölgede F.graminieaum izolatlarının buğday ekim alanlarında daha yaygın olduğu gözlenmiştir.
English In this study it has been investigated to identify of F.graminiarum and F.culmorum based on mycological and molecular analysis in Cukurova region and selection of resistance to wheat head blight used embriogenic calli of three genotypes of wheat (Triticum aestivum L.) Adana 99, Seri 82 and Genç 99. Wheat head blight caused F.graminearum and F.culmorum were identified on wheat fields and F.graminearum was the most widely distributed species in Cukurova region. Immature and mature embrio and immature seeds of wheat varieties used in vitro selection were cultured on MS medium containing 2,4-D to initiate embriogenic calli The frequency of callus induction was 45-92% from immature embrio, 0-21% from immature seeds, and 57-70% from mature embrio. The wheat varieties calli were cultured on modified MS medium containing different concentration of Fusarium culture filtrate and fusaric acid for 4 weeks and the best selective effect was obtained at concentration of 30% culture filtrate and 0,30 mM fusaric acid.
German In dieser Studie wurde es für die Ermittlung von F.graminiarum und F.culmorum auf der Grundlage von mykologischen und molekulare Analyse Cukurova Region untersucht und Auswahl des Widerstandes gegen Weizen Kopf bLicht verwendet Embriogenic Calli der drei Genotypen von Weizen (Triticum Aestivum L.) Adana-99, Seri 82 und Genç 99. Weizen Kopf Knollenfäule verursacht F.graminearum und F.culmorum wurden auf Weizenfeldern und F.graminearum war die am meisten verbreiteten Arten Cukurova-Region. Unreife und Reife Embrio und unreifen Samen von Weizen-Sorten, die in Vitro-Auswahl verwendet wurden kultiviert auf MS Medium mit 2,4-D zu initiieren Embriogenic Calli die Häufigkeit der Hornhaut Induktion war 45-92 % von unreifen Embrio, 0 bis 21 % aus unreifen Samen und 57-70 % aus Reifen Embrio. Die Weizen-Sorten-Calli wurden modifizierte MS mittlere mit unterschiedlichen Konzentration der Fusarium Kultur Filtrat und Fusarinsaeure für 4 Wochen kultiviert und die besten selektive Wirkung wurde bei einer Konzentration von 30 % Kultur Filtrat und 0,30 mM Fusarinsaeure erreicht.
