Descriptions : Turkish Bu çalışmada on bir turunçgil anacının in vitro koşullarda geliştirilen bitkilerinin kök segmentleri, kotiledon, epikotil, yaprak ve embriyo eksplantları kullanılarak embriyogenik kallus (EK) eldesi hedeflenmiştir. Eksplantlar farklı besi ortamlarında kültüre alınmıştır. Çalışma sonucunda, embriyo eksplantlarından elde edilen kalluslardan en yüksek embryogenik kallus oluşum oranı 2,4-D (2 mg/l)+BAP (0.5 mg/l) içeren besi ortamında Troyer sitranjı (Citrus sinensis (l) Osb. X Poncirus trifoliata (L) Raf) anacında %80 EK oluşumu gözlenmiştir. Epikotil, yaprak ve kotiledon kültürlerinin her üçünde de en iyi besi ortamı 2,4-D (2 mg/l)+KİN (0.5 mg/l)+NAA (2 mg/l) içeren denemede EK oluşumu gözlenmiştir. Epikotil eksplantı denemesinde, en iyi EK oluşumu Rubidoux (Poncirus trifoliata (L) Raf.) üç yapraklı genotipinde görülürken, yaprak eksplantında Severinia buxifolia genotipi en iyi EK olarak bulunmuştur. Kotiledon eksplantı denemesinde de en iyi EK oluşumu Troyer sitranjı genotipinde rastlanmıştır. Kök eksplantı denemesinde en yüksek EK Gou Tou turuncu (Citrus aurantium L.) genotipinde görülmüştür. En iyi besi ortamı ise 2,4-D (2 mg/l) + KİN (0.5 mg/l) olmuştur.
English In this research, eleven citrus rootstocks were cultured to obtain embryogenic callus (EC) from root, cotyledon, epicotyls and leaf segments of in vitro plantlets and embryo culture of seeds. Explants were cultured in different plant growth media. The results showed that the highest EC obtained from embryo culture of Troyer rootstock in the plant medium including 2, 4-D (2 mg/l)+BAP (0.5 mg/l) with 80 %. In the all epicotyls, leaf and cotyledon culture, the best plant media in the formation of EC was 2,4-D (2 mg/l)+KİN (0.5 mg/l)+NAA (2 mg/l). In the experiment of epicotyls explants, the best formation of EC was observed in Rubidoux (Poncirus trifoliata (L) Raf.), where as the best genotype for EC production was Severinia buxifolia in leaf explant. By using cotyledons explant, Troyer citrange also showed the best EC formation. In the experiment of root explants, the highest EC production was obtained from Gou Tou sour orange (Citrus aurantium L.). The best plant medium was 2,4- D (2 mg/l) + KİN (0.5 mg/l).
German In dieser Untersuchung wurden elf citrus Unterlagsreben kultiviert, um embryogenic Callus (EG) von der Wurzel, Keimblatt, Epicotyls und Blatt Segmente von in-vitro-Jungpflanzen und Embryo Kultur der Samen. Explantaten wurden in verschiedenen Pflanzen-Wachstum-Media kultiviert. Die Ergebnisse zeigten, dass die höchste EG aus Embryo Kultur der Troyer Wurzelstock in der Pflanze Medium einschließlich 2, 4D (2 mg/l) + BAP (0,5 mg/l) mit 80 % gewonnen. In der alle Epicotyls war Blatt und Keimblatt Kultur, besten Pflanze Medien in der Bildung der EG 2,4-D (2 mg/l), KİN (0,5 mg/l) + NAA (2 mg/l). Im Experiment von Epicotyls Explantaten wurde die beste Bildung der EG in Rubidoux (Poncirus Trifoliata (L) Raf.), beobachtet dort als der beste Genotyp für EG Produktion Severinia Buxifolia in Blatt Explant war. Mithilfe der Keimblätter Explant Troyer Citrange zeigte auch die besten EG-Bildung. Im Experiment der Wurzel Explantaten wurde die höchste EG-Produktion von Gou Tou saure Orange (Citrus Aurantium L.) erhalten. Das beste Werk Medium war 2,4-D (2 mg/l) + KİN (0,5 mg/l).
