Descriptions : Turkish Bu çalışma, çekirdeksiz karpuzun temel üretim materyali olan tetraploid karpuz bitkisi elde edilmesine yönelik olarak planlanmıştır. Çalışmada, Crimson Sweet karpuz çeşidi kullanılmış ve iki in vivo ile iki in vitro yöntemin tetraploid bitki elde edilmesine etkileri araştırılmıştır. In vivo yöntemlerin birincisinde 7 farklı dozda (% 0.0, 0.1, 0.2, 0.3, 0.4, 0.5 ve 0.6) hazırlanan kolhisin çözeltisi, ilk gerçek yaprak oluşumu aşamasında bitkinin büyüme ucuna damlatılmış; diğer yöntemde ise % 0.0, % 0. 5 ve % 1.0 dozlarında hazırlanan kolhisin çözeltisine serada büyütülen diploid karpuz bitkilerinin büyüme ucu daldırılmıştır. In vitro'da yapılan denemelerde 4 farklı BA dozu (0, 1, 2 ve 4 mg/l) içeren MS kültür ortamlarında kotiledon yapraklarından rejenerasyon sağlanmış, diğer yöntemde ise in vitro diploid mikro çeliklere kolhisinin 3 dozu (% 0.0, 0.5 ve 1. 0) uygulanmıştır. Araştırma bulgularına göre Crimson Sweet karpuz çeşidinde tetraploid bitki elde edilmesine yönelik en başarılı uygulamanın in vitro'da 4 mg/l BA içeren MS ortamından elde edilen rejenerasyon ile gerçekleştiği ve bu uygulamada katlama oranının % 60.4 olduğu tespit edilmiştir. İkinci başarılı metot olarak in vitro kolhisin uygulamasında (% 0. 5 dozu) % 36.4 oranında tetraploid bitki elde edilmiştir. Denemede yer alan in vivo yöntemlerin katlanma başarısı % 5 oranında kalmış ve yeterli bulunmamıştır. In vivo ve in vitro yöntemler sonucu elde edilen bitkilerin ploidi seviyeleri flow sitometri analiz yöntemi ile belirlenmiştir. Tetraploid bitki teşhisinde morfolojik ve sitolojik gözlemlerden de yararlanılmıştır.
English This study was planned to produce tetraploid watermelon plant which is the main production material of seedless watermelon. In this study watermelon cultivar Crimson Sweet was used and the effects of 2 in vivo and 2 in vitro methods on the obtention of tetraploid plant were investigated. In the first in vivo method, 7 different colchine solutions (0.0 %, 0.1 %, 0.2 %, 0.3 %, 0.4 %, 0.5 % ve 0.6 %) were applied by droping to the apical growing point at the first true leaf stage in the other technique, the apical growing points of the greenhouse grown plants were immersed in to (0.0 %, 0.5 % and 1.0 %) colchine solution. In the first experiment of in vitro methods, the cotyledons were regenerated in MS medium containing 4 different concentrations (0, 1, 2, 4 mg/l) of BA were added. In the other technique, 3 different colchicine solution (0.0 %, 0.5 % and 1.0 %) were applied to the in vitro diploidmicrocuttings. According to the results, the most succesful method to obtain tetraploid plants of C. Sweet watermelon cultivar was the regeneration in MS medium containing 4 mg/l BA was added and the duplication rate was 60.4 %. The second successful method was the in vitro colchicine application (0.5 %) with rate of 36.4 % tetraploid plants. The duplication rate of in vivo methods was 5 % that was not found enough successful. The ploidy level of the plants which obtained by in vivo and in vitro methods was analysed with flow cytometry. Morphological and cytological observations were also done to determine tetraploid plants.
German Diese Studie wurde tetraploide Wassermelone Pflanzen zu produzieren, das die Hauptproduktion kernlose Wassermelone sind geplant. In dieser Studie Wassermelone Sorte Crimson Sweet diente und die Auswirkungen von 2 in-vivo und in vitro Methoden 2 über die Erlangung von tetraploide Pflanzen untersucht wurden. In der ersten in-vivo-Methode 7 verschiedene Colchine-Lösungen (0.0 %, 0,1 %, 0,2 %, 0,3 %, 0,4 %, 0,5 % Ve 0,6 %) wurden durch Ablegen auf der apikalen wachsenden Punkt in der ersten Phase der echten Blattes in die andere Technik angewendet, die apikale wachsenden Punkte des Gewächshauses Pflanzen angebaut wurden mitten in (0,0 %, 0,5 % und 1,0 %) Colchine-Lösung. Im ersten Experiment von in-vitro-Methoden, die Keimblätter wurden aufbereitet in MS mittlere mit 4 verschiedene Konzentrationen (0, 1, 2, 4 mg/l) der BA wurden hinzugefügt. Beim anderen Verfahren wurden 3 verschiedene Colchicin-Lösung (0.0 %, 0,5 % und 1,0 %) auf die in-vitro-Diploidmicrocuttings angewendet. Nach den Ergebnissen die meisten erfolgreichen Methode tetraploide Pflanzen von C. Sweet Wassermelone Sorte zu erhalten war die Regeneration in MS Medium mit 4 mg/l, die BA hinzugefügt wurde, und die Vervielfältigung betrug 60,4 %. Die zweite erfolgreiche Methode wurde die in-vitro-Colchicin-Anwendung (0,5 %) mit 36,4 % tetraploide Pflanzen. Die Vervielfältigung der in-vivo-Methoden betrug 5 %, die nicht genug erfolgreich gefunden wurde. Die Ploidie-Ebene der Pflanzen, die durch in-vivo und in vitro Methoden erlangt wurde mit Durchflusszytometrie analysiert. Morphologische und zytologische Beobachtungen wurden auch durchgeführt, um festzustellen, tetraploide Pflanzen.
