Descriptions : Turkish Yapılan bu çalışma ile Spirulina platensis (cyanophyta)'e ait serin esteraz geni iki farklı primer dizileri kullanılarak PCR teknigi ile çogaltılmıştır. Çogaltılan gen, primer dizilerinden kaynaklanan farklılıga baglı olarak, Escherichia coli vektörü olan pBR325 plazmid DNA'sına iki farklı şekilde takılmış ve böylece S. Platensis için entegratif vektörler geliştirilmiştir. Yeni entegratif vektörler pBR325SE1 ve pBR325SE2 olarak isimlendirilmiştir. pBR325SE1 ve pBR325SE2 kompetent E. Coli XL1 Blue MRF' hücrelerine transfer edilirken, pBR325SE1 S. platensis'e de elektroporasyon ile aktarılmıştır. S. platensis/pBR325SE1 kloramfenikol (cml) içeren Spirulina ortamında ürerken, plazmid aktarılmamış olan S. platensis beklenildigi üzere aynı besi ortamında ürememiş, buna karşılık cml içermeyen Spirulina ortamında üremiştir. Bakteriyofaj T4'e ait lizozim geni pBR325SE2 entegratif vektörüne takılarak pBR325SL vektörü geliştirilmiştir. pBR325SL önce kompetent E. coli XL1 Blue MRF, sonrada S. platensis hücrelerine aktarılmıştır. S. platensis/pBR325SL cml içeren Spirulina ortamında ürerken, plazmid aktarılmamış olan S. Platensis beklenildigi gibi aynı besi ortamında ürememiş, buna karşılık cml içermeyen Spirulina ortamında üremiştir. pBR325SL vektöründe lizozim geninin varlıgı E. Coli hücrelerinden izole edilen vektörlerde PCR ile gösterilirken, S. platensis/pBR325SL hücrelerinin DNA izolasyonu yapılacak kadar ürememesinden dolayı, lizozimin varlıgı gösterilememiştir.
English In this study, serin esterase gene from Spirulina platensis amplified by PCR using two different primer sequences. The amplified gene cloned into the pBR325 vector in two different way, depending on differences between primers, thus, two entegrative vectors for S. platensis were constructed. The new entegrative vectors were identified as pBR325SE1 and pBR325SE2 respectively. While pBR325SE1 and pBR325SE2 were transferred into competent E. coli, besides this, pBR325SE1 was electroporated into S. platensis. Recombinant S. platensis/pBR325SE1 were grown in the Spirulina medium with chloramphenicol (cml) while there wasn't any non-transformant Spirulina in the same medium but intensive non-transformant growth was observed in the medium without cml. Bacteriophage T4 lysosyme gene was cloned into pBR325SE2 and the new vector harbouring lysozyme gene was identified as pBR325SL. pBR325SL was transferred into competent E. coli and S. platensis respectively. Recombinant S. platensis/pBR325SL observed in the Spirulina medium with cml while there was no non-transformant Spirulina in the same medium but intensive non-transformant growth was observed in the medium without cml. While the presence of lysozyme gene was demonstrated in pBR325SL isolated from E. coli by PCR, but it couldn't be demonstrated in S. platensis/pBR325SL because of insufficient Spirulina in the medium with cml growth for DNA isolation.
German In dieser Studie, Serin-Esterase-gen von Spirulina Platensis verstärkt durch PCR unter Verwendung von zwei verschiedenen Primer-Sequenzen. Das verstärkte gen geklont in der pBR325-Vektor zwei anders, je nach Unterschiede zwischen Primer, also zwei Entegrative Vektoren für S. Platensis gebaut wurden. Die neue Entegrative-Vektoren wurden jeweils als pBR325SE1 und pBR325SE2 identifiziert. Während pBR325SE1 pBR325SE2 in kompetente E. Coli, darüber hinaus überführt wurden, war pBR325SE1 Electroporated in S. Platensis. Rekombinante S. Platensis/pBR325SE1 wurden in das Spirulina-Medium mit Chloramphenicol (Cml) zugenommen, aber es gab nicht-Transformant-Spirulina im gleichen Medium aber intensive nicht-Transformant Wachstum wurde im Medium ohne FZ Bakteriophage T4 Lysosyme gen geklont wurde, in pBR325SE2 und neuen Vector Beherbergung Lysozym-Gen wurde identifiziert als pBR325SL beobachtet. pBR325SL wurde in kompetente E. coli und S. Platensis bzw. übertragen. Rekombinante S. Platensis/pBR325SL in das Spirulina-Medium mit Cml zu beobachten, während im gleichen Medium gab es keine nicht-Transformant-Spirulina aber intensive nicht-Transformant Wachstum im Medium ohne FZ verzeichneten Während das Vorhandensein von Lysozym-gen in pBR325SL demonstriert wurde von E. Coli durch PCR isoliert, aber es konnte nicht in S. Platensis/pBR325SL wegen unzureichender Spirulina im Medium mit Cml Wachstum für die DNA-Isolierung nachgewiesen werden.
