Descriptions : Turkish Bu arastırmada, laktik asit bakterilerinden (LAB) elde edilen serbest hücre (SH) ekstraktının patojen mikroorganizmaların (PB) gelişimi ve iki farklı amino asit (histidin ve tirozin) sıvı besiyerinde amonyak ve biyojenik amin (BA) üretimi üzerindeki etkileri belirlenmiştir. SH ekstraktlarının antimikrobiyal etskisi agar difüzyon yöntemi kullanılarak test edilmiştir. Bu çalışmada 4 adet LAB üyesi (Leuconostoc mesenterodies subsp. cremoris, Pediococcus acidophilus, Lc. Lactis subsp. lactis, Streptococcus thermophilus) ve 4 adet patojen suşu (Listeria monocytogenes, Staphylococcus aureus, Escherichia coli, ve Salmonella parathypi A) kullanılmıştır. Bu çalışma sonucunda, SH ekstraktları bakteri gelişimini önemli düzeyde inhibe etmiştir (P<0.05). En yüksek inhibisyon etki Staph. aureus için Strep.thermophilus'dan ve S. paratyphi A için Leu. mes. subsp. cremoris ve Lc. Lactis subsp. lactis elde edilen SH ekstraklarından gözlenmiştir (12.33 mm). Bakteriler tarafından üretilen amonyak (AMN) ve BA üretimi bakımından önemli farklılıklar gözlenmiştir (P<0.05). Histidin dekarboksil sıvısında (HDS) S. paratyphi, L.monocytogenes, Staph.aureus ve E.coli tarafından histamin üretimi sırasıyla 10387.537, 722.01, 7330.66 ve 11163.53 mg/L bulunmuştur. Tirozin dekarboksil sıvısında (TDS) ise bu bakteriler tarafından tiramin üretimi 367.54 ile 5955.86 mg/L arasında değişkenlik göstermiştir. AMN ve BA üretiminde LAB üyelerinden elde edilen SH ekstraktının etkisi kullanılan dekarboksil besiyeri ve bakteri türüne göre değişkenlik göstermesine karşın, SH ekstraktı PB tarafından üretilen AMN ve BA miktarını genellikle önemli düzeyde düşürmüştür (P<0.05). Ancak, LAB üyelerinin SH ekstraktının PB tarafından HDS'de tiramin, TDS'da ise dopamin ve serotonin üretiminde artırıcı özellik gösterdiği bulunmuştur
English In this study, the functions of cell free (CF) extract from lactic acid bacteria (LAB) on the growth and biogenic amine (BA) production of food borne pathogens (FBP) in two amino acid (histidine and tyrosine) decarboxylase broth were investigated. The antimicrobial functions of CF extract on the growth of FBP were tested using agar diffusion method. Four LAB (Leuconostoc mesenterodies subsp. cremoris, Pediococcus acidophilus, Lc. lactis subsp. lactis, Streptococcus thermophilus and four pathogenic bacteria strains (Listeria monocytogenes, Staphylococcus aureus, Escherichia coli and Salmonella parathypi A were used. The result of the study showed that CF extract significantly inhibited growth of FBP (P<0.05). The highest inhibition effects were found by Strep. thermophilus for Staph. aureus, by Leu. mes. subsp. cremoris and Lc. lactis subsp. lactis for S. paratyphi A (12.33 mm inhibition zone diameter). There was significant differences in ammonia and BA production among the bacteria species (P<0.05). In histidine decarboxylase broth (HDB), histamine production by S. paratyphi A, L. monocytogenes, Staph. aureus and E. coli were 10387.54, 722.01, 7330.66 and 11163.53 mg/L, respectively. In tyrosine decarboxylase broth (TDB), tyramine production by these bacteria ranged from 367.54 to 5955.86 mg/L. CF extract from LAB generally inhibited AMN and BA production by FBP (P<0.05), although the functions of CF extract varied depending on not only decarboxylase broth but also bacteria strains. However, CF extract from LAB had stimulation effect on tyramine production in HDB and dopamine and serotonin production in TDB by FBP
German In dieser Studie extrahieren Sie die Funktionen der Zelle frei (CF) aus Milchsäure Bakterien (LAB) auf das Wachstum und die Biogene Amine (BA)-Produktion von Lebensmittel übertragenen Erreger (FBP) in zwei Aminosäuren-Dekarboxylase Brühe (Histidin und Tyrosin) untersucht wurden. Die antimikrobiellen CF-Extrakt das Wachstum des FBP-Funktionen wurden getestet mit Agar-Diffusion-Methode. Vier LAB (Leuconostoc Mesenterodies subsp. Cremoris, Pediococcus acidophilus, Lc. Lactis subsp. Lactis, Streptococcus Thermophilus und vier pathogenen Bakterien-Stämme (Listeria Monocytogenes, Staphylococcus Aureus, Escherichia coli und Salmonellen Parathypi A dienten. Das Ergebnis der Studie zeigte, dass CF-Extrakt deutlich Wachstum der FBP gehemmt (P < 0,05). Die höchste Hemmung-Effekte wurden durch Streptokokken gefunden. Thermophilus für Staph. Aureus, von Leu. MES. subsp. Cremoris und Lc. Lactis subsp. Lactis für S. Paratyphi A (12,33 mm Hemmung Zone Durchmesser). Gab es erhebliche Unterschiede in Ammoniak und BA-Produktion unter den Bakterienarten (P < 0,05). Histidin-Decarboxylase-Bouillon (HDB), Histamin-Produktion von S. Paratyphi A, L. Monocytogenes, Staph. Aureus und E. Coli waren 10387.54, 722.01, 7330.66 und 11163,53 mg/L. In Tyrosin-Decarboxylase-Bouillon (TDB), Tyramin Produktion durch diese Bakterien reichten von 367.54 bis 5955,86 mg/L. CF Auszug aus LAB generell gehemmt AMN und BA Produktion von FBP (P < 0,05), obwohl die Funktionen des CF-Extrakt je nach nicht nur Decarboxylase-Bouillon, sondern auch Bakterienstämme variiert. Allerdings hatte CF Auszug aus LAB Impulswirkung auf Tyramin Produktion in HDB und Dopamin und Serotonin in TDB von FBP
